The finding that mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC) has resulted in considerable interest in the understanding of the mechanism of DNA mismatch repair. Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes mutS and mutL were demonstrated at high frequency in HNPCC and were shown to be associated with microsatellite instability. A member of the mismatch repair family, GTBP (also designated MSH6), is an MSH2-related protein that binds to DNA containing G/T mismatches. Findings suggest that the mismatch-binding factor in human cells is composed of a heterodimer of GTBP and MSH2. CHD4/Mi-2b is a component of the nucleosome remodeling and deacetylase (NuRD) complex which is a multi-subunit protein complex containing both histone deacetylase and nucleosome-dependent ATPase subunits. Current models predict that this complex function primarily in transcriptional repression. Accumulating evidence indicates that NuRD may regulate the transcription of specific genes by interacting with specific transcriptional factors. In addition, the NuRD complex may also participate in genome-wide transcriptional regulation through an association with histone tails. Recognizes a protein of 55 kDa, which is identified as GLUT-1. Glucose transporters are integral membrane glycoproteins involved in transporting glucose into most cells. There are many types of glucose transport carrier proteins, designated as Glut-1 to Glut-12. Glut-1 is a major glucose transporter in the mammalian blood-brain barrier. It is expressed in high density on the membranes of human erythrocytes and the brain capillaries that comprise the blood-brain barrier. Glut-1 is expressed at variable levels in many human tissues. Overexpression of Glut-1 has been linked to tumor progression or poor survival of patients with carcinomas of the colon, breast, cervical, lung, bladder and mesothelioma. Glut-1 is a sensitive and specific marker for the differentiation of malignant mesothelioma (positive) from reactive mesothelium (negative). Recognizes the L1 or Calprotectin molecule, an intra-cytoplasmic antigen comprising of a 12 kDa alpha chain and a 14 kDa beta chain. Calprotectin comprises 60% of the cytoplasmic protein fraction of circulating polymorphonuclear granulocytes and is also found in monocytes, macrophages and ileal tissue eosinophils. Peripheral blood monocytes carry the antigen extra- and intracellularly, neutrophils only intracellularly. Calprotectin has antibacterial, antifungal, immunomodulating and antiproliferative effects. Besides this it is a potent chemotactic factor for neutrophils. Plasma concentrations are elevated in diseases associated with increased neutrophil activity, like inflammatory bowel disease. Granulocytes terminate their existence after transmigration through the intestinal wall. Therefore calprotectin is also detectable in feces. Recognizes a protein of 94 kDa, which is identified as the glucose-regulated protein 94 (grp94) and also tumor rejection antigen (gp96). Grp94 shows a high degree of sequence homology with the heat shock protein 90 (hsp90). This MAb is highly specific to grp94 and shows minimal cross-reaction with other members of the HSP90 family. Grp s are a class of proteins unresponsive to heat shock and are induced by glucose deprivation. Napsin is a pepsin-like aspartic proteinase connected with maturation of surfactant protein B.There are two closely related napsins, napsin A and napsin B. Napsin A is expressed as a single chain protein. Immunohistochemical studies revealed high expression levels of napsin A in human lung and kidney but low expression in spleen. Napsin A is expressed in type II pneumocytes and in adenocarcinomas of lung. The high specificity expression of napsin A in adenocarcinomas of lung is useful to distinguish primary lung adenocarcinomas from adenocarcinomas of other organs.

Recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. This antibody reacts with a carbohydrate determinant, identified as CDw78 (Workshop V; Code CDw78.2). Epitope recognized by this MAb and Leu21 is the same. It is present on some immature and some peripheral blood B cells, and some B cell lines. Its expression is increased after B-cell activation in vitro. CDw78 is also found on tissue macrophages and on epithelial cells, but not on T cells, NK cells, monocytes, granulocytes and myeloid tissues. It appears that CDw78 is a determinant on a major histocompatibility complex (MHC) class II subpopulation.
